Review of Noncancer Risk Assessment: Application of Benchmark Dose Methods
نویسنده
چکیده
The overall goal of this project is to evaluate risk assessment methods traditionally used for noncancer health risks and to compare them with new approaches. Initially this report gives a brief economic rationale for the impact of prevention of noncancer health effects. This is shown using figures for years of potential life lost which reveal that noncancer health effects, such as birth defects, are on the same national economic magnitude as cancer and heart disease. Traditional approaches for assessing these noncancer risks are discussed. These methods include identification of no observed adverse effect levels (NOAELs). Reference dose (RfD) calculation or setting of acceptable daily intake (ADI) values is achieved by dividing the NOAEL values by uncertainty and/or modifying factors. These factors represent a default approach to

Application of transcriptional benchmark dose values in quantitative cancer and noncancer risk assessment.
The traditional approach for estimating noncancer and cancer reference values in quantitative chemical risk assessment is time and resource intensive. The extent and nature of the studies required under the traditional approach has limited the number of chemicals with published risk assessments. In this study, female mice were exposed for 13 weeks to multiple concentrations of five chemicals th...
متن کاملSYMPOSIUM OVERVIEW Biologically Based, Quantitative Risk Assessment of Neurotoxicants
The need for biologically based, quantitative risk assessment procedures for noncancer endpoints such as neurotoxicity has been discussed in reports by the United States Congress (Office of Technology Assessment, OTA), National Research Council (NRC), and a federal coordinating council. According to OTA, current attention and resources allocated to health risk assessment research are inadequate...
متن کاملApplicable risk assessment methods in occupational and environmental exposure to nanoparticles - a narrative review
Nanoparticles (NPs) are a heterogeneous group of materials that have various applications, and their risk assessment is an essential condition. This study aimed to review the applicable risk assessment methods in occupational and environmental exposures to NPs. A literature search for articles published since 2005 in Web of Knowledge, Scopus, PubMed, Science Direct, and Google Scholar, using ap...
متن کاملRisk characterization framework for noncancer end points.
The nature of both indoor air exposures and noncancer end points present significant issues for risk characterization. Noncancer end points are multidimensional, affecting various organs, and are assumed to have thresholds. Symptoms also vary in severity within a population. In addition to the complexity of noncancer risk assessment, indoor air exposures are typified by the presence of complex ...
متن کاملEvaluating noncancer effects of trichloroethylene: dosimetry, mode of action, and risk assessment.
Alternatives for developing chronic exposure limits for noncancer effects of trichloroethylene (TCE) were evaluated. These alternatives were organized within a framework for dose-response assessment--exposure:dosimetry (pharmacokinetics):mode of action (pharmacodynamics): response. This framework provides a consistent structure within which to make scientific judgments about available informati...
